Im 7. Jh. v.Chr. wurde das Territorium des späteren Sinope von den Milesern besiedelt und zu einer vitalen und wohlhabenden Kolonie ausgebaut. 183 v.Chr. nahm Pharnakes I. Sinope ein und erhob sie zur Hauptstadt des Königreiches Pontos. Die Blütezeit Sinopes endete mit dem Sieg des Pompeius Magnus über Mithridates VI., den König von Pontus, im Jahre 64 v.Chr.
